How to Read a Certificate of Analysis

A field-by-field walkthrough of the sections that appear on a peptide certificate of analysis and what each result does and does not establish.

What a certificate of analysis is

A certificate of analysis (COA) is a laboratory document that records the analytical results obtained for one specific lot of material. It is a snapshot of what an analytical laboratory measured on a defined sample, on a defined date, using defined methods.

A COA describes the tested lot only. It does not describe a product line, a brand, or any lot that was manufactured before or after the tested one, which is why lot matching is the first step in reading one.

Fields to locate first

  • Compound name and any synonym used by the testing laboratory.
  • Lot or batch identifier, which must match the identifier printed on the vial.
  • Analysis date and, when present, the report issue date.
  • Analytical methods applied, commonly HPLC for purity and mass spectrometry for identity.
  • Reported result for each method, including the acceptance criterion when the laboratory states one.
  • Appearance and physical form of the sampled material.
  • Name of the laboratory that performed the analysis.

Reading the purity result

A chromatographic purity figure expresses the area of the main peak as a percentage of total integrated peak area under the stated method. It is a relative measure within the conditions of that method, not an absolute statement about every possible impurity.

A purity value therefore answers a narrow question: under this method, how much of the detected material eluted as the main component. It does not by itself establish identity, sterility, endotoxin status, or fitness for any particular application.

Reading the identity result

Mass spectrometry data on a COA typically report an observed mass that is compared against the calculated mass for the intended molecule. Agreement within the method tolerance supports the identity assignment.

Where a COA reports only purity and no identity method, the document supports a purity conclusion alone. Treat the absent field as absent rather than assuming an unstated result.

Limits of a COA

  • A COA does not convert a research material into an approved drug product.
  • A COA does not state that material is safe or suitable for human or veterinary use.
  • Sterility and endotoxin conclusions require their own named tests; absence of those tests means no such claim is supported.
  • Results apply to the sampled lot and to the sampling point in time.

What is the first field to check on a COA?
The lot or batch identifier. If it does not match the identifier on the vial in hand, the document does not describe that material.
Does a high purity percentage mean a material is sterile?
No. Purity is a chromatographic measure of composition. Sterility is a separate microbiological determination and requires its own stated test result.
Why do two laboratories report slightly different purity values?
Chromatographic purity depends on column chemistry, gradient, detector wavelength, and integration settings. Different validated methods can return slightly different area percentages for the same lot.
Does a COA make a compound approved for human use?
No. A COA is an analytical record. It carries no regulatory approval and no authorization for human or veterinary administration.
